Although both characters contribute to the 50 headshot count... so if they have both unlocked the gun, you can safely switch between them to keep the count going until one of them gets the 50th.

Because it is tied to your account, the answer is most likely no.

You can get the pistol with your second character, but the $250k is limited to one per account, therefore there will be no more.

Although both players contribute to the 50 headshot count, you can safely switch between them to keep the count rolling until one of them gets the 50th.
