So let's say friend A and i start the DDH with just us, with him as host.

Can I start doing the heist with friend B with friend B as host?

Or will that mess up those challenges?

To put it another way, can you work on two heists, hosted by two different people, off and on? Or does that mess up those challenges?

It will mess them up. Loyalty is with the same people with the same host. In-order as it implies, is by playing them in order.
