I love GTA Online and hope they support that for a long time, but I would love something along the lines of GTA IV's episodes. With three different characters, or at least maybe one or two of the original three changing. I think that's a lot of work, though, and for all the manpower it would take they could be working on a new, more profitable title, rather than DLC. I have a feeling that DLC will be strictly Online for this game. Anything to keep the currency valuable, so people keep paying real money for it.
