It's a more subtle version of the 69 meme. Agreed. As Rockstar are notorious with adding 69 jokes to GTA games, the Route 68 is a subtle meme to it. Sort of like "Right, we've used 69 Easter eggs a lot, so lets stop it. Add 68 then " Plus, it's a road. I don't want my mates asking for my location and me telling them that I am heading south on the Route 69. It sounds silly and the humour of it would dry out.
