Depending on the image, you will need specific image editing software like Photoshop or GIMP (free) to create your image on multiple layers, then simply delete the layer which contains the background and the remainder of your image will be transparent. Then you must save as a .PNG or a .GIF as other formats do not support transparency and will automatically fill in the background.

 

For already 'flattened' images, the process is more complex. There's a whole range of techniques to delete the background of an image, the most common simply being to use the Eraser tool in your image editing software, though the results of this method aren't the best. You could use the selection (or 'magic wand') tool to select your background and delete it, but this is only really effective if the background contains many soild colours. The most reliable method is masking, of which there are many tutorials online as well as the GFX Requests & Tutorials archive, which is similar to the eraser except it doesn't ruin your image, if you delete something important you can simply restore it, but this method takes the longest. So as you can see there are many ways to get the same result.
