First of all, don't overfill it because A) it makes it hard to open and B) it is well documented that it "eats" vehicles. If it's not full, you should be able to simply drive up to it and it should open. The Doherty garage is usable from the start of the game, although under normal circumstances you would get a four-star wanted level for being in forbidden territory at that time. The interior you see in the cutscenes does not load inside the building in the regular world, though. "You tell me exactly what you want, and I'll explain to you very carefully why it cannot be."
