So I'm playing GTA SA again and noticed that the score for this mission is 2500 (PS2) but have seen in some YT videos that the minimum score for this one on the PS2 is 4000. Can someone confirm the differences between v3.0 (the one that I own) and the earlier releases?

That has been discussed to death. Later versions of the game have decreased difficulty in particular missions like Life's a Beach and Supply Lines. Nothing new.

V1/2 PS2 version of Supply Lines' bar works as a timer and can result very difficult to complete.

Wrong.

 

PS2 v1 = Life's A Beach requires 4000 points or better to complete the mission, and Supply Lines still burning the fuel even if you aren't pressing X to accelerate.

 

PS2 v2 as "The Introduction", "Greatest Hits" or later revisions = Life's A Beach requires 2500 points or better to complete the mission, and Supply Lines difficulty it was downgraded to make it less hard and the fuel "bug" has been fixed. *This latter applies to later ports as PC, Xbox and Mobile. PS4 is the PS2 version running on an embedded PS2 emulator and of course, is v2.
