I really love just taking a drive about the map, sometimes cruising at leisurely speeds, and sometimes speeding recklessly, avoiding collisions with vehicles and other obstacles (or occasionally colliding into oncoming traffic with a heavier vehicle intentionally). This will be very fun in GTA V, because of the range of environments and the size of the map. The days after I get the game will be especially fun, because that's when I'll be discovering the map. I'll be more familiar with it after that point, but hey, I still often take a drive around GTA IV's map to this day. I'll also enjoy walking about the world on foot, to explore the nooks and crannies that I might otherwise miss while driving around.
And secondly, rampaging is also a favorite activity of mine in GTA and games like it. Shooting people seems like it'll be more satisfying than ever in V, with improved mechanics, better 'gore', and a large variety of weapons, and Michael's ability to slow down time while shooting will make for some fun times watching people fall with blood splattering in slow motion (while shooting them as they're still falling). It's going to be a lot of sadistic fun.
But ultimately what I look forward to most is experiencing everything the game has to offer. For instance, I'm imagining putting on a scuba diving suit, getting into a plane, flying around for a bit, then bailing out over water, sky diving into the ocean (or aiming to fall into a smaller body of water for extra challenge), and exploring the depths. I bet there's way crazier combinations that I'll think of when I get the game, particularly with combining the multiple protagonists. So like Fisciletti said, my favorite activity in GTA V for a while will be simply playing the game and exploring its possibilities. But I think as I become more familiar with the game, I'll settle on driving around and rampaging.

Driving and exploring will be so much fun in a game on this scale and especially with multiplayer.
